Male Contraceptive Initiative Invests In Hormone-Free Birth Control Pill For Men
In a significant stride towards reproductive health equity, the Male Contraceptive Initiative (MCI) has announced a $500,000 investment in YourChoice Therapeutics. This San Francisco-based biotech startup is on the brink of introducing the first hormone-free male birth control pill, YCT-529, marking a pivotal moment in the development of male contraceptives. The initiative's funding supports the ongoing Phase 1a clinical trial of YCT-529, which has successfully enrolled 16 male volunteers as of December 2023.
Heather Vahdat, Executive Director of MCI, highlighted the importance of this development during Women's History Month, emphasizing the need to address gender inequities in contraceptive responsibilities. Historically, the onus of pregnancy prevention has predominantly fallen on women, with a plethora of contraceptive options like pills, patches, and devices. In contrast, men's options have been severely limited. The introduction of a male birth control pill could significantly alter societal norms, promoting shared responsibility in pregnancy prevention and paving the way towards reproductive equality.

YCT-529 operates by inhibiting sperm production through blocking access to retinoic acid in male testes. This mechanism has been validated by multiple independent studies and represents a novel approach to male contraception that avoids the hormonal side effects associated with female contraceptives. Preliminary NIH-funded preclinical studies have shown YCT-529 to be 99% effective in preventing pregnancies in mice and 100% reversible without any side effects.
Akash Bakshi, co-founder and CEO of YourChoice Therapeutics, expressed gratitude for MCI's support and emphasized the groundbreaking nature of YCT-529 as the first hormone-free birth control candidate for men undergoing human studies. Bakshi pointed out that while the pill for women has been revolutionary, it has also subjected them to hormonal side effects for over six decades. YCT-529 offers an opportunity for men to actively participate in contraceptive practices without the adverse effects linked to testosterone suppression found in previous male contraceptive attempts.
Research indicates a strong consensus among men and women that contraception should be a shared responsibility. However, past efforts to develop a male birth control pill have been hindered by reliance on testosterone suppression, which can lead to undesirable side effects such as increased cardiovascular disease risk, weight gain, acne, mood changes, and decreased libido. YCT-529's hormone-free approach could thus provide a more appealing option for men seeking effective and reversible birth control methods.
The development of YCT-529 is based on decades of research into the role of vitamin A in male fertility. By targeting the retinoic acid receptor-alpha (RAR-a), YCT-529 prevents both the production and release of sperm cells in the testes. This innovative approach was developed in collaboration with Dr. Gunda Georg, a renowned medicinal chemist and recipient of one of the largest NIH-funded Contraceptive Centers grants.
The ongoing Phase 1a clinical trial for YCT-529 is being conducted at Quotient Sciences in the UK, with further details available on clinicaltrials.gov under identifier NCT06094283.
